泥蚶体液和肌肉提取液凝集素的初步研究
A Preliminary Study on Lectins of Humoral and Muscular Extract from Tegillarca granosa
【摘要】 利用14种天然或经酶修饰的动物及人红细胞来测定泥蚶的体液和肌肉提取液的凝集活力,并且进行糖(200 mmol·L-1)抑制、热稳定性、pH及Ca2+ 影响试验.结果显示:泥蚶体液和肌肉提取液对14种红细胞均有凝集作用.体液对兔红细胞的凝集作用可被乳糖和半乳糖所抑制,其凝集活性依赖于Ca2+,在pH7.0较稳定,经90 ℃保温10 min后活性完全消失;肌肉提取液的凝集活性不被实验所用的10种糖所抑制,在pH6.0~7.0,温度为60 ℃~90 ℃范围内均具较高活性,其凝集活性也依赖于Ca2+.推测:泥蚶的体液与肌肉提取液均含有凝集素,且种类不同.
【Abstract】 Humor and muscular extract from Tegillarca granosa were detected for hemagglutination with 14 natural and enzyme-treated erythrocytes of human and animals, in addition, glyco-inhibiting action(200 mmol·L-1), thermostability and influence of pH and Ca2+ on the activity of the two kinds of samples were tested.The results showed that both humor and muscular extract could agglutinate 14 kinds of erythrocytes,but their agglutination titers were different.Agglutination activity of humor to rabbit erythrocytes was inhibited by lactose and D-galactose. The hemagglutination activity of humor depended on Ca2+,and it showed the highest hemagglutination activity in pH 7.0,temperature 60 ℃.The agglutination activity of muscular extract to rabbit erythrocytes was not inhibited by the following ten saccharides, i.e. sorbose, lactose, xylose, maltose, L-Rhamnose, D-mannose, D-fructose, D-galactose, D-glucose and sucrose. It had highest hemagglutination activity in pH range of 6.07.0,temperature range of 60 ℃90 ℃, its hemagglutination activity also depended on Ca2+.The results supposed that both humor and muscle of Tegillarca granosa contain lectin,and they are different in nature.
