含复方中药大鼠血清对人卵巢癌新鲜实体瘤细胞增殖影响的研究
Effects of medicated rat serum of traditional Chinese herb decoction on proliferation of ovarian cancer fresh solid tumor cells
【摘要】 目的探讨复方中药治疗卵巢癌的作用机制。方法应用中药血清药理学的实验方法,利用大鼠制备含药血清,采用细胞排染法和3H掺入法观测含药血清对39例卵巢癌新鲜实体瘤细胞增殖的作用。结果细胞排染法检测低、中、高浓度含药血清对卵巢癌新鲜实体瘤细胞生长的抑制率在上皮性肿瘤中分别为(13.36±2.38)%、(27.68±3.76)%和(43.24±4.54)%,在性索间质肿瘤中分别为(9.53±2.16)%、(21.32±3.09)%和(38.27±3.52)%,在生殖细胞肿瘤中分别为(8.88±1.98)%、(19.08±3.62)%和(37.25±3.46)%。在各病理类型中,随含药血清浓度的增加,抑制率增高,P<0.05。在不同浓度含药血清中,对卵巢上皮性癌细胞增殖的抑制率均明显高于对生殖细胞和性索间质细胞肿瘤细胞增殖的抑制率,P<0.05。经空白血清及低、中、高浓度含药血清处理的卵巢癌新鲜实体瘤,3H掺入法检测的的cpm值在上皮性肿瘤中分别为526.24±36.68、445.63±36.72、378.21±37.82和302.16±32.36;在性索间质肿瘤中分别为530.63±36.72、508.87±45.86、468.93±46.27和403.11±37.54;在生殖细胞肿瘤中分别为523.21±37.82、501.11±44.67、447.22±46.58和381.79±37.2。同一种卵巢肿瘤病理类型中,含药血清处理组细胞增殖cpm值明显低于空白血清组,P<0.05;而且不同浓度含药血清组cpm值比较差异有统计学意义,P<0.05。不同浓度含药血清组中,卵巢上皮性肿瘤细胞的cpm值明显低于生殖细胞肿瘤和性索间质肿瘤细胞的cpm值,P<0.05。结论实验所用复方中药可抑制人卵巢癌新鲜实体瘤细胞的增殖。
【Abstract】 OBJECTIVE:To explore the treatment mechanism of traditional Chinese herb decoction on ovarian cancer. METHODS: The effects of the medicated serum on 39 cases of ovarian tumor fresh solid tumors cell were investigated with Trypan blue dye exclusion assay and 3H-TdR incorporation. RESULTS: 1) Trypan blue dye exclusion assay showed that tie inhibition rates of cell proliferation were (13.36±2.38)%, (27.68±3.76)% and (43.24±4.54)% in epitheial ovarian tumor cells, (9.53±2.16)%, (21.32±3.09)% and (38.27±3.52)% in ovarian gonadal sex cord stromal tumor cells, and (8.88±1.98)%, (19.08±3.62)% and (37.25±3.46)% in ovarian germ cell tumor cells treated with rat serum contained different concentrations of traditional Chinese herbs respectively, P<0.05. As the increase of traditional Chinese herb concentration, the inhibition rates of cell proliferation significantly increased correspondingly in the three ovarin tumer cells, P<0.05. The result of assay of 3H-TdR incorporation suggested that the cpm were 526.24±36.68, 445.63±36.72, 378.21±37.82 and 302.16±32.36 in epithelia ovarian tumor cells, 530.63±36.72, 508.87±45.86, 468.93±46.27 and 403.11±37.54 in ovarian gonadal sex cord stromal tumor cells, and 523.21±37.82, 501.11±44.67, 447.22±46.58 and 381.79±37.2 in ovarian germ cell tumor cells treated with rat serum contained different concentrations of traditional Chinese herb respectively. For one type of ovarian tumor, the cpm of tumor cells in traditional Chinese herb groups was significantly lower than that in the control group, P<0.05 , Furthermore, a significant difference was found among all traditional Chinese herb groups, P<0.05. At the same concentration of traditional Chinese herb, the cpm in epithelia ovarian tumor cells was significantly lower than that in ovarian germ cell tumor cells and ovarian gonadal sex cord stromal tumor cells, P<0.05 . CONCLUSION: The traditional Chinese herbs decoction can inhibit proliferation of ovarian tumor fresh solid tumor cells.
