原发性结直肠癌组织中乳清酸磷酸核糖转移酶的表达及临床意义

【摘要】 目的:探讨人原发性结直肠癌组 织中乳清酸磷酸核糖转移酶(orotatephos phoribosyltransferase,OPRT)的表达及其临 床意义。方法:应用RT PCR方法测定61 例原发性结直肠癌手术切除标本及正常对 照组织OPRT的基因表达水平,分析OPRT 与原发性结直肠癌的临床关系。结果:结 直肠癌肿瘤组织中OPRT mRNA表达水平 是非肿瘤组织中的4.38倍,P=0.0001, OPRT mRNA的表达量在黏液腺癌中最低, 在低分化腺癌中最高。OPRT mRNA在肿 瘤组织和非肿瘤组织中的基因表达水平存 在正相关关系,rs=0.55,P=0.0001。肿 瘤组织中的OPRT mRNA/非肿瘤组织组织 中的OPRT mRNA的比值(T0/N0)与淋巴 结转移(rs=0.36,P=0.005)、脉管转移(rs =0.26,P=0.041)和组织学分化(rs= 0.33,P=0.009)具有相关性。结论:与非 肿瘤组织相比,肿瘤组织中OPRT mRNA有 较高表达水平;OPRT在肿瘤组织和非肿瘤 组织中的基因表达水平存在正相关关系。 OPRT mRNA的表达量在黏液腺癌中最低, 在低分化腺癌中最高,淋巴结转移、脉管转 移和组织学分化与T0/N0降低有关。

【Abstract】 OBJECTIVE:To study the expression of OPRT and its clinical significance in human primary colorectal cancer. METHODS: RT-PCR was used to determine the expression levels of OPRT gene in primary tumors (T) and normal tissues (N) in 61 patients with primary colorectal carcinoma. The relationships between OPRT gene expression and its clinicopathological parameters were analyzed. RESULTS: The expression of OPRT-mRNA was approximately 4.38-folds higher in colorectal cancer tissues than that of non-tumor ones,P=0.000 1. The lowest expression of OPRT-mRNA was found in mucous adenocarcinoma while the highest level in poorly-differentiated adenocarcinoma. Expression of OPRT in tumor tissues showed a positive correlation with one of paired non-tumor tissues,r s=0.55,P=0.000 1. There were correlations between tumor tissues’ OPRT-mRNA expression/non-tumor tissues’OPRT-mRNA expression ratio (T 0/N 0) and metastasis to the lymph nodes,r s=0.36,P=0.005, small vessel invasion (r s=0.26,P=0.041) and differentiation (r s=-0.33,P=0.009). CONCLUSIONS: The expression levels of OPRT-mRNA are higher in colorectal cancer tissues compared with ones of non-tumor tissues. Expression of OPRT in tumor tissues shows a positive correlation with one of paired non-tumor tissues. The lowest expression of OPRT-mRNA is found in mucous adenocarcinoma while the highest level in poorly-differentiated adenocarcinoma. T 0/N 0 ratio is correlated to metastasis to the lymph nodes, small vessel invasion and differentiation.
