苏木醇提取物治疗实验性重症肌无力小鼠的临床疗效及对其T淋巴细胞功能的影响

【摘要】 目的采用中药苏木醇提取物治疗实验性重症肌无力(EAMG)小鼠,观察其临床疗效及T淋巴细胞功能变化,探讨其防治重症肌无力的作用机制。方法采用乙酰胆碱受体(AChR)加等量福氏佐剂多次免疫小鼠,复制EAMG小鼠模型,EAMG治疗组小鼠胃内注入苏木醇提取物,其余对照组给予等量0.9%氯化钠溶液,观察治疗前、后小鼠体重及临床症状变化,并于治疗30d后处死小鼠,检测其T淋巴细胞功能。结果EAMG治疗组小鼠体重较治疗前显著增加,临床症状明显缓解(P<0.05);T淋巴细胞增殖反应受到明显抑制,与EAMG对照组间差别有显著性意义(P<0.05)。结论苏木能明显缓解重症肌无力症状,促进神经功能恢复,部分机制是通过下调T淋巴细胞功能,从而抑制N2AchR抗原诱导的特异性免疫反应而起到治疗作用。

【Abstract】 Objective To explore the therapeutic effects and the mechanism of Caespalpinia Sappan L.when it was treated with the myasthenia gravis(MG),the clinical function and T lymphocyte competence were observed in experiment autoimmune myasthenia gravis(EAMG) rats with the ethanol extracts of Caespalpinia Sappan L.Methods The EAMG model in rats was induced by immunizing the rats for more times with the mixture of acetylcholine receptor ( AChR ) and complete Freuds adjuvant.The ethanol extracts of Caesalpinia Sappan L were given to the EAMG treatment group from the stomach and the equal physiological salt water (0.9%) to the other control group every day.The rats′ weight and the clinical score were observed and the T lymphocyte transformable competence was tested after killing the rats thirty days later.Results In EAMG treatment group,the rats′ weight were increased more obviously than those before the treatment,the clinical score was decreased clearly (P<0.05);the T lymphocyte transformable competence was suppressed in the EAMG treatment group.Compared with EAMG control group there was significant difference(P<0.05).Conclusion The Caesalpinia Sappan L.is able to ameliorate muscular weakness and improve the neurological function in EAMG rat.Some mechanisms can suppress the T lymphocyte transformable competence and restrain specific immune reaction by N2AChR antigen.
