【摘要】 目的研究顺铂作用于卵巢癌CAOV3细胞后,端粒酶活性、细胞凋亡和细胞周期变化与细胞耐药性的关系。方法用顺铂处理卵巢癌CAOV3细胞,端粒酶重复序列扩增(TRAP)-银染法测定端粒酶活性,流式细胞仪观察细胞凋亡和细胞周期的改变。结果顺铂(DDP)处理卵巢癌CAOV3细胞后,CAOV3细胞端粒酶活性受到抑制,细胞周期受阻于G2/M期,未见细胞凋亡峰改变。结论CAOV3细胞对顺铂的耐药性通过多因素起作用,DDP对端粒酶活性具有抑制作用,可以改变细胞周期,但对细胞凋亡无明显影响。更多还原

【Abstract】 Objective To investigate the drug resistance to cisplatin of the ovarian cancer CAOV3 line and the relationships between the drug resistance and telomerase activity,apoptosis,cell cycle.Methods CAOV 3 cells were treated with cisplatin.Telomerase activities were assayed by the method of telomeric repeat amplification protocol (TRAP)-silver straining.The change of cell cycle and apoptosis were analyzed by flow cytometer.Results It was found that telomerase activity of CAOV 3 cells inhibition and cell cycle accumulation in G2/M phase without apoptosis phase during the treatment.Conclusion The drug resistance to cisplatin of ovarian cancer CAOV 3 line is effected by several agent,which is not changed directly the inhabition of cisplatin to CAOV 3 cells or induce the apoptosis of cell CAOV 3 line.The apoptosis induced by cisplatin closely related to cell cycle.更多还原
