离心造粒法制备醋氯芬酸微丸

【摘要】 目的制备醋氯芬酸微丸。方法采用离心造粒法制备醋氯芬酸微丸,考察工艺因素对微丸收率的影响,并采用正交设计法对影响微丸收率的主要因素进行筛选优化。结果在成丸的工艺过程中,主机转速、喷浆泵转速、空气流量、滚圆时间、供粉转速对微丸收率均有影响,经正交试验优化得微丸成丸的最佳工艺参数为:醋氯芬酸与微晶纤维素(MCC)的质量比为10∶1,黏合剂为质量分数是3%的羟丙基甲基纤维素(HPMC,5MPa·s)水溶液。主机转速为20Hz,喷浆泵转速为30 r·min-1,空气流量为0.5 m3·h-1,滚圆时间为4 min,供粉转速为30 r·min-1。结论在优化条件下采用离心造粒法可制得表面较为光滑、圆整度较高的醋氯芬酸微丸,450~595μm微丸的收率可达w=78.3%。

【Abstract】 Objective To prepare aceclofenac pellet(AFP). Methods The AFP was prepared by using a centrifugal granulator. The effect of technology on the yield of pellets was investigated. The optimal conditions of technology were selected by the orthogonal design. Results The rotating rate of plate, the spraying rate of binder solution pump, the rate of airflow, the time of spheroidization and feeding rate of powder were found to have influences on pelletizing. The process parameters established by the orthogonal design were as follows: the ratio of core pellets and MCC was 10:1, the adhesive agent was 3% HPMC solution. The rotating rate of plate wad 20Hz, the rotating rate of spray solution pump was 33r·min-1, the rate of airflow was 0.5m3·h-1, the time of spheroidization and the rotating rate of powder feed machine was 30r·min-1. Conclusion Under the optimal conditions, the pellets prepared by using centrifugal granulator have perfect shape and surface characteristics and the yield of 450~595 μm pellets could reach to 78.3%.
