UTI和丹参在鼠肝脏缺血再灌注中对肝窦内皮细胞损伤保护效应的实验研究
Protective effects of Ulinastatin and Danshen on liver sinusoidal endothelial cell damage during ischemia and reperfusion of rats
【摘要】 目的探讨UTI和丹参在肝脏缺血再灌注中对肝窦内皮细胞损伤的保护作用。方法将大鼠肝脏缺血再灌注模型30只大鼠分为5组,A组为空白对照组(n=3),B组为缺血再灌注组(n=7),C组为UTI组(n=6),D组为丹参组(n=6),E组为UTI+丹参组(n=8)。除A组外,在缺血30min再灌注120min后分别取血检测AST、ALT、LDH、HA及ET,并取肝组织行光镜和电镜观察。结果C组、D组较B组各指标明显下降(P<0.01),E组各指标较C组或D组均有下降,(P<0.05)。光镜和电镜观察形态学损伤程度,B组最重,C组、D组次之,E组最轻。结论UTI、丹参在缺血再灌注中能有效减轻对肝窦内皮细胞的损伤,二药合用有协同作用。
【Abstract】 Objective:To investigate the protective effects of Ulinastatin(Urinary trypsin inhibitor UTI)and Danshen on liver sinusoidal endothelial cell(SEC)during ischemia and reperfusion of rats.Methods:Thirty male Wistar rats were randomly divided into five groups group A,Nil control group(n=3);group B,ischemia reperfusion(I/R)group(n=7);group C,UTI treated group(n=6),group D,Danshen treated group(n=6),group E,UIT+Danshen treated group (n=8),which were studied on the basis of the model pre established successfully.The latter 4 groups were reperfused 120 minutes after 30 minutes hepatic ischemia,Serum alanine aminotransferase (AST),aspartate aminotransferase(ALT),Lactic dehydrogenase(LDH),hyaluronic acid(HA) and plasma endothelin(ET)were respectively measured in each group,the test data were analyzed by SPSS10.0 and SASv6.2.In addition,liver samples were obtained for light and electronic microscopy study.Results:The AST,ALT,LDH,HA and ET levels in group C and group D were significantly lower than those in group B(P<0.01).their levels of means in group E were lower than those in group c or group D,too.However only ET level is significant(P<0.05),according to statistics analysis.The morphological injury changes were mild in group E compared with group C or group D,and group B was the severest.Conclusion:This experiment suggests that UTI and Danshen could prevent SEC from injury during hepatic ischemia and reperfusion in rat liver,it is better when two medicines were.administratived concomitantly.
