Apelin激活大鼠主动脉阳离子氨基酸转运体的表达

【摘要】 目的研究Apelin对大鼠离体主动脉阳离子氨基酸转运体(CAT)和诱导型一氧化氮合酶(iNOS)基因表达的影响。方法体外血管薄片孵育,测定孵育液中亚硝酸盐(NO2-)含量、RT-PCR方法测定阳离子氨基酸转运体CAT-1、CAT-2B和iNOS mRNA表达水平。结果大鼠主动脉孵育3h,Apelin(10-9-10-7mol/L)呈浓度依赖地刺激血管NO2-生成增加(33%、46%和69%,均P<0.01)。Apelin刺激血管L-Arg转运体CAT-1和CAT-2B mR-NA基因的表达上调,分别较对照组增加1.11倍和1.28倍(均P<0.01),而iNOS mRNA的表达并无明显改变(P>0.05)。结论Apelin激活血管组织阳离子氨基酸转运体CAT-1和CAT-2B基因表达,其变化可能参与了NO的释放从而引起血管的扩张。

【Abstract】 Objective In this present study,we assess the gene expression of cationic amino acid transporter-1(CAT-1),CAT-2B and inducible nitric oxide syntheses(iNOS) in Apelin-stimulated rat thoracic aorta.Methods The aortic slices were incubated in vitro.Twelve rats were randomized to either a normal group or apelin group.The aortic NO production was determined the medium nitrite content.Semi-quantitative RT-PCR was used to determine the mRNA concentrations of iNOS andCAT-1 and CAT-2B in aorta.Results After incubation of aorta for 3 hour,apelin(10-9-10-7mol/L) increased nitrite production(33%,46% and 69%,all P<0.01) in dose dependent manner.Apelin-induced CAT-1 and CAT-2B mRNA up-regulation and iNOS mRNA unchanged in aorta.Conclusion We provide the first evidence to illustrate that Apelin induces the transcription of high-affinity CAT-1 and CAT-2B in aorta.Transcription of CAT-1 and CAT-2B correlates well with aortic NO biosynthesis.
