蛛网膜下腔出血后兔脑血管肌钙蛋白mRNA表达的研究

【摘要】 目的观察蛛网膜下腔出血(SAH)后兔脑血管平滑肌细胞内肌钙蛋白(calponin)mRNA水平表达的变化规律。方法将25只雄性新西兰白兔随机分为对照组及SAH组,SAH组又分为第1、4、7、14天组,共5组,每组5只兔。采用二次注血法建立SAH后脑血管痉挛的动物模型,以血管造影检测兔基底动脉的血管直径变化,用逆转录-聚合酶链反应(RT-PCR)方法检测alponinmRNA表达变化。结果SAH组第1、4、7、14天组calponinmRNA表达分别为0.34±0.08,0.32±0.08,0.28±0.04和0.15±0.04;对照组calponinmRNA为0.37±0.02,与SAH各组比较,差异有显著意义(P<0.05)。结论calponinmRNA在SAH后逐渐降低,可能与脑血管痉挛的发生、发展有关。

【Abstract】 Objective To investigate the expression of calponin mRNA in cerebral vascular smooth muscle cells during vasospasm after subarachnoid hemorrhage. Methods Rabbit vasospasm models after subarachnoid hemorrhage were induced by twice injections of blood into the cisterna magna.Diameter of basilar arteries were measured on day 1,4,7,14 after injection by angiography and the expression of calponin mRNA was detected by reverse transcription-polymetrase chain reaction (RT-PCR)at the same timing. Results The expression of calponin mRNA was significantly decreased in cerebral vascular smooth muscle cells on day 1(0.34 ± 0.08),day 4(0.32 ± 0.08),day 7(0.28 ± 0.04) and day 14(0.15 ± 0.04) compared with controls(0.37 ± 0.02). Conclusion The expression of calponin mRNA in cerebral vascular smooth muscle cells decreases progressively after subarachnoid hemorrgage in rabbits, which may contribute to the persistent contraction of smooth muscle.
