大鼠脑缺血-再灌注损伤期脑温度变化规律的研究

【摘要】 目的探讨大鼠局部脑缺血再灌注脑温度变化的规律。方法将30只SD大鼠随机分为缺血再灌注组和假手术对照组,每组各15只。对缺血再灌注组大鼠使用线栓法制作大脑中动脉闭塞模型(MCAO),缺血3h再灌注48h。持续监测缺血后1h和再灌注后1h大鼠纹状体和脑皮质的温度变化,同时监测肛温。保持室温在22~25℃,湿度在60%。结果脑缺血再灌注组在缺血早期,纹状体温度下降1.3℃,脑皮质温度下降1.5℃,肛温变化不明显;再灌注后,纹状体和脑皮质温度反跳性升高0.5~0.8℃,其中脑皮质温度快速升至(36.3±0.6)℃,纹状体温度升至(36.8±0.8)℃。肛温变化仍不明显。假手术组脑温度相对稳定。结论缺血期脑温度先下降后缓慢升高,再灌注时脑温度反跳式升高后稍有下降,随后缓慢升高。

【Abstract】 Objective To study the changes of brain temperature during cerebral ischemia-reperfusion injury in rats. Methods 30 rats were equally devided into stroke group and control group. In the stroke group, brain ischemia was produced by thread occlusion of the middle cerebral artery for 3 hours and then reperfused for 48 hours. In the control group, only sham surgery was done. Brain temperatures was monitored ipsilaterally in the area supplied by the MCA for 1hour both during ischemia and reperfusion. Body temperature was also measured frequently through the rectum both during ischemia and reperfusion. The ambient temperature and humidity was controlled strictly to keep constant. Results Brain temperature dropped approximately 1.3℃ [from (37.1 ± 0.5)℃ to (35.8 ± 0.8)℃ in stratum, and from (36.5 ± 0.6)℃ to (35.0 ± 0.8)℃]in the cortex during earlier period of ischemia, and increased 0.5~0.8℃[from (36.5 ± 0.6)℃ to (36.8 ± 0.8)℃ in the stratum, and from (35.8 ± 0.6)℃ to (36.3 ± 0.6)℃]in the cortex during reperfusion in the stroke group. The rectum temperature of the rats changed lightly during both ischemia and reperfusion period. The brain temperature and rectum temperature of the rats in control group were relatively steady. Conclusion This study suggest that the brain temperature drop during ischemia and rebound during reperfusion period, and support the theory that mild hypothermia may be helpful to alleviate ischemia-reperfusion injury of the brain.
