【摘要】 以4个石榴(PunicagranatumL )品种为实验材料,用SDSⅠ,SDSⅡ,CTABⅠ,CTABⅡ4种方法提取石榴叶片DNA 结果表明,SDS法提取得率为500～600μg·g-1,但OD值偏低,为1 50～1 70;RAPD分析无扩增;CTAB法所得DNA得率为200～500μg·g-1,OD值为1 80～1 90;RAPD分析扩增带清晰CTABⅠ和CTABⅡ相比,操作简单、省时,得率高,综合考虑CTABⅠ是石榴叶片DNA提取的最佳方法更多还原

【Abstract】 4 methods were tested to extract Punica granatum DNA from leaves using 4 cultivars as material.The results showed that SDS methods had a higher extraction rate of 500～600μg·g^-1,but a lower OD value of 1.50～1.70 without amplifying bands for RAPD analysis.The extraction rate was 200～500μg·g^-1 of CTAB methods higher,OD value was 1.80～ 1.90 with clear amplifying bands for RAPD analysis.Compared with CTABⅡ,CTABⅠ was simpler, time saving and had higher extraction rate.All things considered CTABⅠ was the best method for extracting Punica granatum L.DNA from leaves.更多还原
