组织芯片及免疫组化技术用于淋巴瘤组织学类型免疫表型和EB病毒关系的观察
Study of Histological Type and Immunophenotype of Lymphomas and Its Relation to Epstein-Barr Virus Infection Using Tissue Chip and Immunohistochemical Technique
【摘要】 目的 :通过观察内蒙古地区淋巴瘤的组织学类型、免疫表型和EB病毒的关系 ,了解内蒙古淋巴瘤病理特点 ,同时探讨组织芯片技术在这方面的使用价值。方法 :采用组织芯片技术结合免疫组织化学和原位杂交对 74例内蒙古地区淋巴瘤进行观察。结果 :本组病例中男性多于女性 (2 8∶1) ;平均患病年龄 4 0 2岁。霍奇金淋巴瘤占所有淋巴瘤的 16 %。霍奇金淋巴瘤均为经典型霍奇金淋巴瘤 ,其中混合细胞型为主要亚型 (6 7% ) ,且与EB病毒关系密切 (6 2 5 % )。非霍奇金淋巴瘤占所有淋巴瘤的 84 % ;结外淋巴瘤的数量近似于结内淋巴瘤 (32和 30例 ) ;B细胞淋巴瘤 (76 % )多于T细胞淋巴瘤 (2 4 % ) ,B细胞淋巴瘤中侵袭性B细胞淋巴瘤占多数 (35例 ,74 % ) ,T细胞淋巴瘤与EB病毒关系密切 (33% ) ,而B细胞淋巴瘤未查出EB病毒感染。结论 :内蒙古地区淋巴瘤的特点近似于我国北方省份的报道 ,而与西方国家报道的结果相差较大。组织芯片在研究淋巴瘤的免疫表型方面较传统切片方法具有快速省时、省力、节约经费、实验误差小及结果易于比较的优点。但是在观察淋巴瘤组织学类型方面由于其组织小 (直径 0 6mm)而存在一定局限性
【Abstract】 Objective:To study pathological features of lymphoma by observing its histological type and immunophenotype and relation to Epstein-Barr Virus(EBV)infection in Inner Mongolia,and evaluate the application of tissue chip technique.Methods:Using tissue chip technique,immunohistochemistry and hybridization in situ methods,74 cases of lymphomas including 12 Hodgkin lymphoma(HL) and 62 non-Hodgkin lymphoma(NHL) were analyzed.Results:The ratio of male to female was 2.8∶1.Of the 74 cases,12(16%) had HL and 62(84%) had NHL.The type of all HL was classical ones.The majority of HL(67%) was mixed cellular type,62.5% of which was closely related to EBV.In NHL,the ratio of nodal to extranodal type was 32∶30.The majority of NHL(76%) was B cellular type,which was not associated with EBV.Aggresive B cellular lymphoma were found in 35 cases(74%).33% of T cellular lymphoma was related to EBV.Conclusion:Features of lymphomas in Inner Mongolia are similar to those reported in Northern China,but differ from those described in western countries.Compared with traditional section-cutting methods,tissue chip technique is fast,simple,economical,less error and easy to compare results though localized in observing histological type for necessary tiny tissue (diameter in 0.6mm).
