应用RAPD技术分析绒山羊DNA的多态性
THE GENEFIC DSVERSITY ANALISIS OF CASHMEVE GOAFS DNA BY RAPD TECHNOLOGY
【摘要】 本实验应用RAPD技术,对辽宁多绒山羊、辽宁绒山羊、内蒙古阿尔巴斯白绒山羊和杂种多绒山羊的22个DNA模板进行了多态性比较和遗传距离分析,根据聚类分析构建了系统发生树状图。36条引物中筛选出11条引物具有多态性扩增片段。得到多态性位点59个,占总位点数的90.77%,分析显示杂种多绒山羊和辽宁绒山羊的亲缘关系较近。
【Abstract】 Random amplified polymorphic DNA(RAPD) analysis was used to estimate genetic diversity and relationship in 22 DNA templates which belong to three Inner Monglia cashmere goat breeds and liaoning cashmere goat breeds.36 primers were used,11primers were screened to generate polymorphic bands.The genetic distance matrix was obtained by proportion of shared fragments.Pendrogram were generated by UPGMA.The results indicated that two Liaoning cashmere goats were clustered together.Kubuqi Cashmere goats was cldosely related to Liaoming cashmere.
