鸡胚破骨细胞的分离培养与鉴定
Isolation,culture and identification of osteoclasts from the chick embryo
【摘要】 从18日龄鸡胚长骨(股骨和胫骨)中机械分离破骨细胞于盖玻片和骨片上培养,以建立鸡胚破骨细胞体外培养方法。结合倒置相差显微镜、抗酒石酸酸性磷酸酶(TRAP)染色、甲苯胺蓝染色及透射电镜(TEM)和扫描电镜(SEM)对其形态和功能进行观察鉴定。结果表明,直接分离的细胞具有典型的破骨细胞的形态特点:属多核大细胞,TRAP染色阳性,能在骨片表面形成吸收陷窝。结论:鸡胚长骨机械分离法可获得一定数量并具有骨吸收活性的破骨细胞。
【Abstract】 To establish the method of isolating the osteoclastic cells from the chick embryo,osteoclasts were isolated from long bones(the femora and tibiae)of the 18-day old chick embryo and then cultured on a glass coverslip or a bone slice.The shape and function of these osteoclasts were observed and identified by phase-contrast microscope,TRAP staining,toluidine blue staining.The results showed that the cells had classic characteristics of osteoclasts:multinucleated giant cells,staining positively for TRAP in cells,forming bone absorptive lacunae on the bone slices.In conclusion,a certain number of osteoclasts could be obtained by the method of mechanical isolation from long bones of the chick embryo and the vitality of the osteoclastic absorption was obvious.
