利用巢式PCR检测玉米细菌性枯萎病菌
Detection of Pantoea stewartii subsp.stewartii by nested-PCR
【摘要】 利用通用引物扩增了玉米细菌性枯萎病菌及其近似种的转录间隔区并进行了序列测定。通过序列比较,设计了一对玉米细菌性枯萎病菌的专化性引物,并对所有参试菌株进行扩增。结果显示:该对引物能从参试的玉米细菌性枯萎病菌中特异性地扩增出1条299bp的条带,而其他参试菌株没有扩增信号。与扩增细菌ITS区域的通用引物结合使用,建立了检测和鉴定玉米细菌性枯萎病菌的巢式PCR技术,其检测灵敏度可达到4个细菌细胞,可以准确、灵敏地检测和鉴定玉米细菌性枯萎病菌。
【Abstract】 Intergenic transcription spacer(ITS)areas of Pantoea stewartii subsp.stewartii(Pss)and related species were amplified with universal primers and sequenced.Based on the conserved sequence determined by alignment using the DNAman software,a pair of specific primers was synthesized.A band of 299 bp was amplified from Pss strains but not from other bacteria tested.Using this pair of specific primers together with universal primers,nested-PCR was developed for monitor Pss which detected less than 4 bacterial cells by this method.
