重组人白细胞介素-10诱导瘤细胞凋亡
Apoptosis of tumor cell induced by recombinant human interleukin-10
【摘要】 目的研究重组人白细胞介素-10诱导瘤细胞凋亡的作用。方法利用基因工程技术制备的rhIL-10,分别作用于体外培养的ScaBer、NCIS446、U937瘤细胞,观察瘤细胞的形态改变,MTT法检测rhIL-10对瘤细胞增殖的抑制作用,电泳观察rhIL-10诱导瘤细胞凋亡的情况,TUNEL法检测瘤细胞凋亡百分率。结果电泳出现典型的DNA“lader”,ScaBer、NCIS446、U937三种瘤细胞的凋亡率为48.5%、45.6%、47.1%,IC50分别为0.043、0.052、0.058mg/mL。结论rhIL-10具有显著抑制瘤细胞生长活性、诱导瘤细胞凋亡的作用。
【Abstract】 Objective To investigate the apoptosis in tumor cell induced by recombinant human interleukin-10 (rhIL-10). Me- thods The rhIL-10 prepared by genetic engineering technology were added to Scaber, ncis446, and U937 tumor cells, respectively, and then the morphologic change of the tumor cells was observed. The proliferation inhibition and apoptosis in tumor cells were measured by MTT assay, TUNEL, and agarose gel electrophoresis. Results Typical DNA "lader" was observed. The apoptosis rates of Scaber, ncis446, and U937 tumor cells were 48.5%, 45.6%, and 47.1%, respectively, while the IC_ 50 of the rhIL-10 were 0.043, 0.052, and 0.058 mg/mL, respectively. Conclusion The data indicate that rhIL-10 has obvious antitumor activity.
