降低棉花胚性愈伤褐化研究
Prevent Browning of Upland Cotton Embryoid Callus
【摘要】 在棉花组织培养中,诱导出的胚性愈伤组织转到继代培养基上时,经常出现褐化现象,需要经过3~4次继代培养才能诱导出正常绿色胚状体。实验通过激素和氮源对诱导绿色胚状体的效应研究,认为氮源配比不合适是褐化现象的主要诱因,与KT、6-BA、IAA等激素关系不明显;经过对MS培养基的改良,一次继代就可以获得绿色胚状体,从而缩短了组织培养周期近2个月。
【Abstract】 In cotton tissue culture,normal green embryoid is usually obtained after 3 or 4 rounds subcultures.But the callus usually turns brown in the subculture from embryogenic callus to somatic embryogenesis.Our research suggests that the brown-turning mainly results from the improper ratio of nitrogen.It also indicated that the content of KT、6-BA and IAA had no significant effect on embryo tissue brown-turning.The normal green embryoid is produced only by once subculture after improvement of MS culture medium,shorten its cycle about two months.
