小鼠生后不同发育阶段睾丸生殖细胞的超微结构研究

【摘要】 目的研究生精细胞、支持细胞、睾丸间质细胞在生后不同发育阶段的超微结构特点,了解雄性生殖细胞发育、分化和成熟的过程,亲代细胞与子代细胞间及3种生殖细胞在发育过程中的相互影响关系。方法分别取生后5天、10天、20天雄性小鼠睾丸,固定后作超薄切片,染色后透射电镜下观察并拍片记录。结果生精小管的细胞组合在不同发育阶段表现不同。生后5天,小管管腔很小,管壁细胞排列呈单层,细胞类型仅有精原细胞和幼稚的支持细胞两种,且均于基膜相贴,睾丸间质细胞不可见。生后10天,小管管腔增大,细胞排列呈2~3层,精原细胞增殖分化,支持细胞、睾丸间质开始发育。生后20天,细胞层数增多,出现各级生精细胞,增殖的子代细胞分布于近腔面,可见胞质间桥和变态发育中的精子;支持细胞、间质细胞发育成熟。结论生精细胞在生精小管中的分布具有规律性,不同发育阶段表现出不同的细胞组合。在发育过程中,间质细胞、支持细胞的结构和功能的成熟先于生精细胞的分化,表明生精细胞的发育分化受间质细胞、支持细胞及基膜的共同参与下完成的。

【Abstract】 Objective: To study the ultrastructure of the spermatogenic、sertoli and leydig cells during postnatal period the process of proliferation, differentiation and maturing of the germ cells and the relationship of these three type cells during different stages. Methods: 5、10 and 20 days after postnatal testes were obtained, fixed, then cut into ultra-thin sections with ultramicrotome, and afterwards observed through the TEM. Results: There were different cell types in different stages. At the 5th day, the lumen of the seminiferous tubule was small, the cells were lined with a single layer, only spermatogonial cells and sertoli cells could be seen, the leydig cells were absent. At the 10th day, the lumen became larger, the cells lined with 2 or 3 layers, spermatogonial cells began to diffentiate, the sertoli and leydig cells began to develop. At the 20th day, the cells layed stratified, the intercellur-bridges and spermatid could be seen, sertoli and leydig cells became to mature. Conclusion: The cells have certain rule in seminiferous tubue; they consist of different cells in different times; sertoli and leydig cells mature ahead of the spermatogonial cells. These findings imply that the spermatogonial cells development depends on the two germ cells, which can enhance the spermatogonial cells proliferation and differentiation during spermatogenesis.
