结核分支杆菌Ag85A/hsp70嵌合DNA疫苗的免疫效应研究
INVESTIGATION OF IMMUNOLOGICAL EFFECT OF CHIMERIC MYCOBACTERIA TUBERCULOSIS AG85A/HSP70 DNA VACCINE IN MICE
【摘要】 目的:比较不同疫苗的免疫效应。方法:昆明鼠50只随机分为5组,即生理盐水组,neo组,BCG组,hsp70与Ag85A联合组,Ag85A/hsp70组,每组10只。两次加强免疫后,检测小鼠血清γ干扰素(IFN-γ)和白细胞介素2(IL-2)的水平。结果:测得各组IFN-γ依次为:471.5±76.8pg/ml、574.4±48.5pg/ml、630.0±60.5pg/ml、815.8±119.4pg/ml、822.8±106.7pg/ml;测得各组IL-2依次为:37.5±5.4pg/ml、39.0±7.9pg/ml、49.3±8.2pg/ml、58.8±14.1pg/ml、73.5±17.6pg/ml。嵌合DNA疫苗组与前4组比较有显著性差异(P<0.01)。联合免疫组与BCG组也有显著性差异(P<0.01)。结论:Ag85A/hsp70嵌合DNA疫苗能刺激更多的免疫活性细胞,免疫效应最强。编码不同抗原基因的多个质粒联合免疫与BCG免疫相比免疫效应更强,也具有较高的免疫保护性。
【Abstract】 Objective: To investigate the immunological effect in mice vaccinated by the chimeric Ag85A/hsp70 DNA vaccine and BCG. Methods: Forty mice were divided into five groups, namely, 0.9% sodium chloride group(n=10), PCI-neo group(n=10), BCG group (n=10), hsp70 and Ag85A group(n=10), and Ag85A/hsp70 group(n=10). The mice were immunized three times with the vaccines. For each of the mouse, the levels of interferon γ(IFN-γ)and interleukin 2 (IL-2) in sera were measured with ELISA kit. Results: The levels of IFN-γand IL-2 in sera of the mice in Ag85A/hsp70 group were significantly higher than that in the other groups(P<0.01). Conclusion: The immunological effect resulting from the chimeric Ag85A/hsp70 DNA vaccine was higher than that from the other.
