实验性脑出血大鼠脑内基质金属蛋白酶-9的表达作用研究

【摘要】 目的 动态观察实验性脑出血大鼠脑内基质金属蛋白酶 9(MMP 9)表达水平的变化及其作用。方法  4 5只大鼠随机分为正常对照组、脑出血模型组和假手术组 ,采用胶原酶复制大鼠脑出血模型 ,通过免疫组织化学和原位杂交技术动态测定不同时间点鼠脑内血肿周围脑组织中MMP 9的表达变化 ;同时另选 4 5只大鼠随机分组同上 ,进行脑含水量的测定 ,以观察MMP 9的表达变化与脑水肿程度的相关性。结果 大鼠正常对照组和假手术组均无MMP 9的蛋白和mRNA表达 ,脑出血模型组在 2 4~ 4 8hMMP 9的蛋白和mRNA表达达高峰 ,以后逐渐下降 ,6~ 96h手术侧其蛋白和mRNA表达量与脑含水量呈正相关。结论 实验性大鼠脑出血早期能诱导血肿周围MMP 9的蛋白和mRNA表达增强 ,可能参与了其脑水肿的形成。

【Abstract】 Objective To observe the change of the expression level of matrix metalloproteinase-9 in the brain after experimental intracerebral hemorrhage(ICH) in rats.Methods The ICH animal models were established by stereotactic injection of Ⅶ collagenase 0.4 IU into the right globus pallidus(GP).Five rats each were sacrificed at 6,24,48 and 96 hours after operation respectively.The expression of MMP-9 protein and gene mRNA in the ICH rat brains were investigated by immunohistochemistry and in situ hybridization. Brain water content(BWC) was determined by comparison of wet and dry weight.Results There were no MMP-9 protein and mRNA positive cells in the brain of NG(normal control group) and SG(sham operation group),but positive cells could be found in ICH model group with peak at 24~48 hours.There was positive correlation between encephaledema on the cerebral hemorrhage side of the rats, especially at the perihematoma site,and expression of MMP-9 protein and mRNA.Conclusion The mechanism of encephaledema after cerebral hemorrhage is complicated and is the result of multiple factors.The increased expression of MMP-9 may be one of them.
