从动物血液中提取基因组DNA方法的比较研究
Comparison of Three Methods of Extracting Genomic DNA from Animal Blood
【摘要】 从血液中提取基因组DNA的传统方法是先得到淋巴细胞,再用蛋白酶K、SDS消化,然后用酚、氯仿抽提,乙醇沉淀.CTAB法提取基因组DNA多应用于植物和微生物,从全血中提取基因组DNA报道甚少.采用改良的CTAB法从全血中提取基因组DNA,并以传统方法和TAKARA Blood Genome DNA Extraction Kit作对照.结果表明:CTAB法可以得到大量的、较完整的基因组DNA,并且纯度达到1.8~2.0,可以满足PCR扩增和限制酶切等实验的要求.
【Abstract】 Traditional methods of extracting genomic DNA from animal blood are as follows.Getting hold of lymphocyte firstly,then digesting it by proteinase K and SDS,extracting by hydroxybenzene and chloroform,depositing by alcohol finally.The method of CTAB,which is extensively applied to extract DNA from plant and microbe,has not been reported to use for animal blood yet.So we used the modified method of CTAB to extract genomic DNA from animal blood,and contrasted it with the method of using TAKARA kit and the traditional method.The results showed that abundant and integrative genome DNA with the purity ranging 1.8~2.0 could be extracted from animal blood by the method of modified CTAB.It is concluded that the genomic DNA extracted with the modified CTAB method can satisfy all requirements of PCR amplification and restriction endonucleases digestion.
