开口毛细管的化学修饰及富集和测定水体中微量蛋白质
FLOW-INJECTION IN-LINE PRECONCENTRATION AND PHOTOMETRIC DETERMINATION OF PROTEIN IN WATER USING CHEMICALLY MODIFIED OPEN CAPILLARY
【摘要】 将聚四氟乙烯毛细管用聚乙烯醇交联涂层后,偶联上活性三嗪染料Cibacron BlueF3GA,制成一种新型的亲合染料Cibacron Blue F3GA修饰的聚四氟乙烯亲合毛细管。以牛血清白蛋白(BSA)作为模型蛋白进行在线富集和检测。方法依据亲合毛细管将低浓度的蛋白质溶液在线预富集后,以KSCN溶液洗脱并形成较高浓度的蛋白质溶液,用偶氮胂K光度法在线定量测定。PVA和Cibacron Blue F3GA的固载量分别为8.6μmol.g-1和0.434 mmol.g-1。毛细管容量为0.58 mg.cm-2BSA。这种分析系统适合微量和痕量蛋白质的在线富集和分析。对常见无机离子及有机物的干扰作了试验,结果表明方法的选择性较好。BSA的线性范围为0~200μg.L-1,检出限为1.5μg.L-1,方法用于水体中蛋白质的测定,回收率在99%~102%之间。
【Abstract】 A novel chemically-modified PTFE affinity open capillary was prepared by cross-linking with PVA coated on the inner surface of the capillary,and coupling with an active triazine dyestuff called cibacron blue F3GA(CBF3GA).The loading capacity of PVA and CBF3GA was 8.6 μmol·g-1 and 0.434 mmol·g-1 respectively.The dynamic column capacity of the capillary was found to be 0.58 mg·cm-2.The prepared PTFE affinity open capillary was tested for its characteristics in the in-line preconcentration of protein using bovine serum albumin(BSA) as a model protein.It was found that the dyestuff,CBF3GA,coupled onto the PTFE/PAV capillary showed a strong adsorption to most of the proteins,and the chemically-modified PTFE/PVA capillary was used satisfactorily in the in-line preconcentration of protein in water.A flow injection system for the on-line analysis of protein was designed.The protein adsorbed on the capillary was desorbed by passing 0.5 mol·L-1 KSCN solution(pH 8) through it,and the protein in the eluate was determined photometrically with arsenazo K as color reagent at the wavelength of 611 nm.By this in-line preconcentration,the concentration of protein was raised to about 500 times.For the determination of BSA.Beer′s law is obeyed in the range of 0200 μg·L-1 with a detection limit of 1.5 μg·L-1.Effects of some co-existing inorganic ions and organic compounds were tested and relatively satisfactory selectivity was obtained.In its application to the determination of two water samples,recoveries among 99% to 102% were obtained.
