双酚A对人胚肝细胞凋亡的影响
Apoptosis Induced by Bisphenol A in Human Embryo Liver L-02 Cell Line
【摘要】 [目的]研究双酚A(BPA)对人胚肝L-02细胞的凋亡的影响。[方法]以MTT来分析不同浓度BPA对人胚肝L-02细胞的细胞存活率的影响;用流式细胞术分析不同浓度BPA对细胞凋亡的影响;通过RT-PCR分析BPA引起的生长抑制、DNA损伤基因GADD45、GADD153、GADD34表达水平的变化。[结果]BPA处理后,L-02细胞的生存率随着BPA浓度的增加而下降,在≥50μmol/L,细胞的存活率显著降低(P<0.01)。随着BPA浓度升高,细胞凋亡率增加,≥50μmol/L可显著升高L-02细胞的凋亡率(P<0.01)。生长抑制DNA损伤基因GADD45、GADD153.GADD34表达水平随着BPA浓度增加表达量也增加,100μmol/LBPA组显著高于正常对照组,但在100μmol/L,BPA+VitC组三种GADD基因表达量均稍降低。[结论]BPA对L-02细胞具有细胞毒性作用,可导致细胞凋亡率升高,与细胞生长调控和DNA损伤相关的GADD45、GADD153、GADD34基因表达升高,VitC可减缓BPA对DNA的损伤作用。
【Abstract】 [Objective] To study apoptosis induced by bisphenol A (BPA) in human embryo liver L-02 cell line. [Methods] L-02 cells were exposed to different concentration of BPA. Survival rate, apoptosis rate, expression level of growth-inhibition and DNA damage genes GADD34, GADD45, GADD153 were measured using MTT,FCM and RT-PCR methods respectively. [Results] L-02 cell survival rate decreased as the concentration of BPA increased.The survival rate was significantly lower than that of the control group when the concentration of BPA≥50 μmol/L(P<0.01) . Apoptosis rate increased as the BPA concentration increased. Apoptosis rate was significantly higher when the concentration ≥50 μmol/L. Expression level of the three kinds of GADD genes increased at the concentration of 50 μmol/L BPA,but decreased at 100 μmol/L BPA+30 μg/L VitC group. [Conclusion] BPA is cytotoxic to L-02 cells. It could cause apoptosis of L-02 cells. Vitamin C can reduce the apoptosis caused by BPA.
