抑癌基因PTEN与MMP-9在肝癌组织中表达与侵袭关系研究

【摘要】 目的探讨抑癌基因PTEN与MMP-9在肝细胞性肝癌(HCC)中表达意义及两者与肝癌侵袭性的关系。方法采用免疫组化(SP法)检测67例HCC及癌旁组织中PTEN与MMP-9表达,并分析两者与肝癌临床病理特征、侵袭性的关系。结果HCC中PTEN表达强度明显弱于癌旁组织,而MMP-9阳性表达强度明显高于癌旁组织,且两者间呈负相关;肝癌组织分化程度、侵袭性与PTEN表达呈负相关,而与MMP-9表达呈正相关。结论PTEN、MMP-9的表达一定程度上反映HCC分化和侵袭性强弱;PTEN缺失可引起MMP-9表达增加,在HCC侵袭、转移中发挥作用。

【Abstract】 Objective To investigate the expression and significance of the tumor suppressor genes PTEN and MMP-9 in hepatocellular carcinoma(HCC),and analyze relationship between the expression and tumor invasion.Methods By using S-P method,the expression of PTEN and MMP-9 in 67 cases of HCC and their pericancerous tissues was detected,and their relationship with clinicopathological characteristics and invasion of HCC was analyzed.Results The positive expression intensity of PTEN in HCC was remarkably weaker than that in pericancerous liver tissues,while the expression intensity of MMP-9 was stronger in HCC than that in pericancerous liver tissues.Moreover,there was a negative relationship between the expression of PTEN and that of MMP-9.Correlation analysis revealed that the expression of PTEN was negatively related to the progress of the tumor pathological differentiation and invasion,whereas that of MMP-9 was entirely contrary.Conclusion The expression of PTEN and MMP-9 may reflect the differentiation and invasive potential of HCC tissues to varying degrees.The deletion of PTEN can result in the overexpression of MMP-9,which may play an important role in the invasion and metastasis of HCC.
