【摘要】 目的检测糖尿病肾病(DN)患者肾小球组织中nephrin的表达,以探讨nephrin在DN患者蛋白尿发生机理中可能的作用。方法免疫荧光染色图像分析及RT-PCR法检测DN组、单纯糖尿病组、单纯血尿组及对照组肾小球组织nephrin的表达水平。结果DN组肾小球组织nephrin的表达量显著低于单纯糖尿病组、单纯血尿组及对照组(P<0.05)。DN组患者肾小球组织中nephrin的mRNA表达量也显著低于单纯糖尿病组、单纯血尿组及对照组(P<0.05)。结论DN患者肾小球组织nephrin的表达减少,可能影响肾小球滤过屏障的通透性,导致蛋白尿的产生。更多还原

【Abstract】 Objective To investigate the possible role of nephrin in the development of proteinu-ria from patients with diabetic nephropathy(DN).Methods Immunofluorescence staining,confocal laserscanning microscopy and RT-PCR analysis were used to investigate the expression of the nephrin in theglomerulus from patients with DN,diabetes mellitus(DM),isolated hematuria and healthy control group.Results In the patients with DN,immunofluorescence staining analysis found that the nephrin expressionin the glomerulus decreased significantly compared with that in DMpatients(P<0.05),isolated hematu-ria patients(P<0.05),and control group(P<0.05).The nephrin mRNA expression in the glomerulusalso decreased significantly compared with that in DM patients(P<0.05),isolated hematuria patients(P<0.05),and control group(P<0.05).Conclusion The dramatic decrease of nephrin expression mightcontribute to the development of proteinuria from patients with DN.更多还原
