DNA分子标记检测家蚕黄海、苏春品种和纯度的研究

【摘要】 利用单粒卵微量DNA抽提法,从家蚕黄海、苏春及其一代杂交种转青蚕卵快速提取出基因组DNA。筛选出的RAPD随机引物S60稳定扩增出2个原种的DNA特异性片段RH-Ⅰ440和RS-Ⅱ1000,克隆和测序后发现分别为463bp和约938bp。利用网上数据库进行序列分析,RH-Ⅰ440的nt2-462与一个家蚕wholegenomeshotgunsequence(WGS)(No:BAAB01096026.1)的nt1644-2104有96%的一致性,其中存在2个Caps。RS-Ⅱ1000的nt3-935与另一个家蚕WGS(No:BAAB01031489.1)的nt1046-1989有97%的一致性,其中存在13个Gaps。RH-Ⅰ440和RS-Ⅱ1000可以作为两个品种RAPD特征DNA片段进行品种和纯度的鉴定。

【Abstract】 The genome DNA of silkworm was extracted from single blue-stage-egg of Huanghai, or Suchun or their hybrids with rapid method. The RH-Ⅰ440 and RS-Ⅱ1000 of specific bands of random amplified polymorphic DNA (RAPD) was amplify traced each from of Huanghai and Suchun variety, with selected random primer of S60. The sequence was 463bp and 938bp respectively after clone and sequence. Then analyze the sequences by courtesy of database on -line. The results showed 96% of nt1644-2104 of one of whole genome shotgun sequence (WGS) has the same sequence as the nt2-462 of RH-Ⅰ440(Genbank accession No:BAAB01096026.1) which include two gaps. Accordingly, the nt3 - 935 of RS-1000 and the nt1046-1989 of the other WGS (Genbank accession No: BAAB01031489. 1) have 97% consistency add to 13 gaps. The conclusion is that RH-Ⅰ440 and RS-Ⅱ1000 were the specific bands of RAPD, could used to identify Bombyx variety and purity of Huanghai and Suchun.
