【摘要】 目的:寻找口腔鳞癌对顺铂耐药的相关基因,以预测临床化疗效果并指导临床化疗。方法:收集口腔鳞癌标本,根据其体外药敏测试(改良MTT法)进行分组,同时用免疫组化法检测这些标本中CyclinDs(CCND1、CCND3)、P鄄gp和GST鄄Pi的表达情况。结果:根据药敏测试结果,最后收集到顺铂耐药标本23例,顺铂敏感标本20例。对免疫组化结果采用SAS5.0统计软件分析,发现只有CCND1和CCND3的表达水平与标本顺铂敏感性有关,CCND1阳性率高往往提示肿瘤细胞对顺铂不敏感。结论:细胞周期因子CyclinDs的表达异常与口腔鳞癌顺铂耐药性相关,可被纳入口腔鳞癌顺铂耐药相关基因表达谱。更多还原

【Abstract】 Objective: Screen and identify those cisplatin-sensitivity related genes and expression profiles in oral squamous cell carcinoma, to help illustrate the underlined reason of drug resistant and direct the chemotherapy in clinic. Methods: Oral squamouse cell samples were collected and divided into two groups according to the results of in vitro modified MTT assay with cisplatin. Immunohistochemical tests were carried out to test the expression status of CCND1, CCND3,GST-Pi and P-gp. Results: According to results of modified MTT assay, the samples were clustered into cisplatin resistant groups(23 Samples) and cisplatin sensitive groups(20 Samples) respectively. There were significant difference in positive rate of CCND1 and CCND3 between these two groups, but not in GST-Pi and P-gp. Conclusion: Expression status of CCND1 and CCND3 could be included in cisplatin -resistant related gene expression profile.更多还原
