MAGE-A1与大肠肿瘤发生发展的关系

【摘要】 目的:研究MAGE-A1在人大肠肿瘤发生、发展过程中的作用,并探讨其与大肠腺癌侵袭和转移的关系.方法:免疫组化S-P法,观察38例正常人大肠粘膜组织、31例大肠腺瘤、83例大肠腺癌组织中MAGE-A1蛋白的表达和定位特点.结果:MAGE-A1蛋白定位于胞浆;在正常大肠粘膜组织中几乎不表达,在大肠肿瘤(大肠腺瘤、大肠腺癌)中有较高的表达;在大肠腺瘤与大肠腺癌中的表达差异无统计学意义;在大肠腺瘤中的表达与患者的性别、年龄、组织学类型、是否复发及癌变之间差异均无显著性;MAGE-A1在大肠腺癌中的表达与患者的性别、年龄、组织学分级、侵袭深度、有无肝转移之间均无统计学意义;与患者有无淋巴结转移之间差异有显著性(P<0·01);且呈正相关关系(P=0·007,rs=0·287).结论:MAGE-A1可能在大肠肿瘤的发生、发展(尤其是淋巴结转移)中起作用.

【Abstract】 Objective: To study the function of MAGE-A1 gene in the carcinogenesis and progression of human colorectal neoplasms and to investigate the correlation with the invasion and metastasis of colorectal adenocarcinoma.Methods: MAGE-A1 was detected at its protein level by SP-immunohistochemistry in 38 normal colorectal mucosal tissues, 31 colorectal adenoma tissues and 83 colorectal adenocarcinoma tissues.Results: The expressions of MAGE-A1 protein was showed in cytoplasms.MA454 almost had no staining in normal colorectal mucosal tissues.In both colorectal neoplasms tissues and normal colorectal mucosal tissues, the expressions of MAGE-A1 protein were significantly decreased (P<0.01). But the expressions of MAGE-A1 protein between colorectal adenoma tissues and colorectal adenocarcinoma tissues had no difference.Furthermore,there was no obvious difference in the expressions of colorectal adenomas in sex, age, 3 histological types of colorectal adenomas,recrudescence and canceration, nor was there apparent difference in sex, age, differentiated grade, depth of wall invasion and liver metastasis.However, there was significantly difference between the expressions of MAGE-A1 in colorectal adenocarcinomas with regional lymph nodes metastasis and those without regional lymph nodes metastasis(P<0.01), and the correlation was positive(P=0.007, r_s=0.287).Conclusion: MAGE-A1 gene may play important roles in occurrence of colorectal neoplasms and progression of colorectal adenocarcinoma, especially in lymph nodes metastasis of colorectal adenocarcinoma.
