非小细胞肺癌中FHIT基因mRNA表达的研究
Study of mRNA Expressions of FHIT Genes in Patients with Non-small Cell Lung Cancer
【摘要】 目的:检测FHIT基因mRNA在非小细胞肺癌(NSCLC)中的表达情况.方法:采用逆转录-聚合酶链反应(RT-PCR)方法对53例NSCLC患者的肺癌进行FHITmRNA表达的检测.结果:53例肺癌标本中FHIT基因mRNA的失表达或异常表达率为64·2%(34/53);非肺癌患者肺组织FHIT基因失表达或异常表达率为0.FHIT基因mRNA的失表达或异常表达率与患者性别、年龄、分期、原发肿瘤大小及有无淋巴结转移均无明显关系(P>0·05),在鳞癌中的失表达或异常表达率为91·3%,在腺癌中的是43·3%,差异有显著性(P<0·05).结论:非小细胞肺癌中存在FHIT基因失表达和转录表达异常,在肺鳞癌中发生率高,FHIT基因的缺失可能是肺癌发生的早期分子事件.
【Abstract】 Objective :To investigate the expressions of FHIT genes in lung cancer samples of patients with non-small cell lung cancer(NSCLC) at mRNA level.Methods: 53 samples of lung cancer from patients with NSCLC were studied to detect mRNA for FHIT gene by using reverse transcription-polymerase chain reaction assay ( RT-PCR).Results: Of the 53 lung cancer samples, the loss and transcript abnormality of FHIT gene was expressed at mRNA level in 64.2%(34/53).In the samples of lung of the patients without lung cancer, the loss and transcript abnormality of FHIT gene was expressed at mRNA level in 0%.It was found that the loss and transcript abnormality of FHIT gene was not related to age, sex, stages of cancer, size of primary tumor and location of cancer.The aberrant rate of FHIT gene in squamous cell carcinoma tissues (91.3%) was significantly higher than that in adenocarcinoma tissues (43.3%).Conclusion: There is loss and transcript abnormality of FHIT gene in NSCLC.FHIT loss and transcript abnormality preferentially occurs in squamous cell carcinomas, FHIT loss may be the early molecular phenomenon of lung cancer.
