【摘要】 目的 :建立并优化人骨髓间充质干细胞 (mesenchymalstemcells,MSCs)分离、消化、传代扩增等培养方法 .方法 :以Percoll (1. 0 73g/mL)、Ficoll (1. 0 77g/mL)和A液 (笔者配制 ) 3种分离介质分离MSCs .传代时以不同浓度胰蛋白酶对MSCs进行消化 .结果 :A液对MSCs的分离效果最佳 ,不同浓度的胰蛋白酶液对MSCs消化能力不同 .结论 :建立了一套稳定、低耗、有效的MSCs培养方法 .更多还原

【Abstract】 Objective:To establish and optimize a method for isolation,digest,subculture and expansion of mesenchymal stem cells (MSCs) from human bone marrow in vitro.Methods:MSCs were isolated with 3 isolation medium,1.073g/mL percoll,1.077g/mL Ficoll and Solution A (author offer).MSCs were digested and subcultured with different concentration trypsase or EDTA.Results:Solution A is the best to isolate MSCs.Different concentration trypsase make different effect for digest of MSCs.Conclusion: A stable,less expensive and efficient method for cultivation of MSCs from human has been established.更多还原
