登革4型病毒广东流行株NS2a-NS2b基因序列分析
SEQUENCE ANALYSIS OF NS2a-NS2b OF DENGUE 4 VIRUSES IN GUANGDONG PROVINCE
【摘要】 目的分析登革 4型病毒 (DV4 )广东株的基因序列 ,比较其同源性 ,追查DV4的地理来源。方法RT -PCR扩增DV4NS2a -NS2b基因片段 ,克隆至PGEMT载体 ,分析其序列。结果1990年分离的 4个毒株在所分析的区段具有相同的序列 ,与DV4H2 4 1、814 6 6 9株、1978年广东株(GD785 6B2 )和 1993年广东株 (GZB5 )的核苷酸 (氨基酸 )同源性分别是 97% (98% )、94 % (96 % )、93% (98% )和 99% (98% )。GD785 6B2株与DV4H2 4 1、814 6 6 9株和 1993年广东株 (GZB5 )的核苷酸 (氨基酸 )同源性分别是 93% (98% )、96 % (97% )和 98% (93% )。结论 1990年引起广东省登革热流行的毒株很可能只有 1个 ,1990年的毒株与 1978年的毒株可能来自不同的疫源地。
【Abstract】 Objective To analyze the gene sequence of Dengue 4 viruses (DV4) in Guangdong Province,to compare their similarity and explore their sources. Methods 398 bp fragment in the NS2a-NS2b region was amplified by RT/PCR, cloned into PGEM T vector, then sequenced.Results The 4 DV4 local strains isolated in Guangdong in 1990 (GD9006A1, GD9033A1, GD9049A2,and GDA63) shared the same sequence in this target region. The nucleotide (amino acid) sequence homology of DV4 isolated in 1990 with DV4H241, 814669, GD7856B2 (isolated in 1978) and GZB5 (isolated in 1993) were 97%(98%)、94%(96%)、93%(98 %)and 99%(98%) respectively. The nucleotide (amino acid) sequence of GD7856B2 with DV4H241, 814669, GZB5 were 93%(98%), 96%(97%), 98%(93%)respectively. Conclusion The dengue fever epidemic in Guangdong in 1990 most possibly resulted from one DV4 strain. The Guangdong DV4 local strains isolated in 1990 and in 1978 came from different sources.
