超声粉碎抗原免疫预防鼠幽门螺杆菌感染的研究
Study on immune mechanism induced by sonicate antigen against Helicobacter pylori infection
【摘要】 目的研究幽门螺杆菌超声粉碎抗原与霍乱毒素B亚单位组成的口服疫苗预防HP感染的作用,探讨其保护性免疫应答机制。方法用HP超声粉碎抗原2mg加CTB10μg作为免疫预防组,另设单纯HP超声粉碎抗原组、单纯CTB组和PBS组为对照组。免疫4周后以活菌攻击,观察各组小鼠的免疫保护率。攻击前后测定血清中IgA、IgG1和IgG2a抗体的变化,小鼠胃黏膜分泌性IgA抗体变化情况。结果免疫预防组小鼠免疫保护率为73.33%,而对照组全部感染HP(P<0.001);攻击前免疫预防组小鼠IgA、IgG1和IgG2a抗体均轻度升高,攻击后IgA、IgG1和IgG2a抗体进一步升高,以IgG2a抗体增高更为明显,IgG1/IgG2a比值从0.67下降至0.59。结论HP超声粉碎抗原加CTB可预防HP感染,并诱导出以Th1反应为主的保护性免疫应答。
【Abstract】 Objective To investigate the effects of the oral vaccine containing helicobacter pylor(HP)sonicate plus cholera toxin B subunits(CTB) in the prevention of HP infection in mice,and study the protective immune response induced by the vaccine. Methods Mice were orally immunized by HP sonicate 2mg plus CTB10μg .Mice immunized with HP sonicate ,CTB alone or PBS were used as control. Four weeks after immunization mice were challenged with HP. The protective rate after challenge,serum anti-HP IgA,IgG1, IgG2a antibodies. and secretory IgA in gastric mucosa in different groups were studied before and after challenge. Results Protective rate of the immunized group was 73.33%. When compared with those of control groups , IgA, IgG1 and IgG2a of the immunized group were increased mildly .After challenge the levels of IgA, IgG1 and IgG2a increased further ,and the immunized group had a significant high level of IgG2a ,so IgG1/IgG2a ratio decreased from 0.67 to 0.59.Conclusion The oral vaccine containing HP sonicate plus CTB is effective in preventing from HP infection in mice ,and can induce a predominant Th1 protective immune response in mice.
