猕猴桃基因组DNA不同提取方法的研究
A Study on Different Methods for Extraction of Kiwifruit Genomic DNA
【摘要】 4种经优化的DNA提取方法即CTAB区室法、SDS区室法、CTAB常规法、SDS常规法提取猕猴桃嫩叶基因组DNA,效果均好。经综合对比分析,本实验认为CTAB常规法是适于AFLP分析最佳的提取方法,因其步骤简单、快捷,得到的基因组DNA降解少、杂质含量少,能获得高质量的DNA模板。通过对所得到的基因组DNA模板进行AFLP分析,得到了清晰的DNA指纹图谱。
【Abstract】 The four optimized methods for extraction of genomic DNA i.e. CTAB subarea method , SDS subarea method , CTAB conventional method and SDS conventional method are practical and efficient. The experiment shows the CTAB conventional method is the best method of applicable to AFLP by compositivly contrastive analysis,because it is simple and quick to obtain high-quality genomic DNA with less elimination and impurity. Using this method very clear AFLP fingerprinting pattern can be obtained.
