拟康氏木霉液态发酵条件的研究

【摘要】 采用单因素和正交试验对筛选出的一株有潜力的生物防治菌——拟康氏木霉Trichoderma pseudokoningii的液态发酵条件进行优化,并进行100L发酵罐中试放大试验的研究。优化后的拟康氏木霉发酵控制参数为:培养基配方为麸皮40g/L,马铃薯50g/L,蔗糖20g/L,KH2PO4 0.5 g/L,MgSO4·7H2O 0.5g/L,CaCl2 0.25g/L;摇瓶发酵培养时间为6d,培养基的初始pH值为6.0- 7.0,发酵温度为28±1℃。通过对拟康氏木霉生长曲线的测定,确定其在发酵罐中培养的时间60h为宜,此时所获得的菌丝体干重为1.2694g/100ml发酵液。本研究结果为高效率、低成本、工业化生产具有生防作用的拟康氏木霉菌丝制剂提供了科学依据。

【Abstract】 The liquid fermentative condition of Trichoderma pseudokoningii were studied through single factor and orthogonal design experiments. 100L fermentation tank pilot study was also conducted. The optimized fermentation control parameters are as follows: culture medium formula is wheat bran 40g/L, potato 50g/L, sucrose 20g/L,KH2PO4 0.5 g/L, MgSO4 · 7H2O 0.5g/L, CaCl2 0.25g/L. The optimum flask fermentation period is 6d. The fermentation temperature and the initial pH-value of culture medium is 28 ±1 ℃ and 6.0-7.0 respectively. The optimum cultivating period is defined as 60h in fermentation tank and the obtained mycelial dry weight reached 1.2694g/100ml of fermentation liquid. The results will provide scientific basis for fast, efficient, and practical industrialization production of Trichoderma pseudokoningii preparation.
