新鲜山羊输卵管液对体细胞克隆和转基因胚胎体外发育的影响
The effect of fresh hydrosalpinx fluid on the in vitro development of goat embryos by somatic cloning and microinjection
【摘要】 用添加不同浓度发情期山羊输卵管液的培养基,对正常单细胞受精卵、注射外源基因后受精卵和细胞核移植后山羊重构卵进行体外培养,探讨其对胚胎体外发育的影响。结果表明:添加20%输卵管液后正常受精卵桑椹胚分裂率为26.1%(11/42),极显著(P<0.01)高于对照组(未添加输卵管液)(7.6%)、50%输卵管液组(4.7%)和100%输卵管液组(0);添加20%输卵管液后,体外培养显微注射转基因受精卵有20.0%(8/40)发育至16-细胞,极显著(P<0.01)高于对照组(6.7%)和50%输卵管液组(4.8%);细胞核移植重构卵在含20%输卵管液的培养基中桑椹胚分裂率为7.3%,与体内培养桑椹胚分裂率(11.1%)差异不显著。在体外培养条件下,添加20%输卵管液可明显提高正常受精卵、转基因受精卵和克隆重构卵的分裂率和发育率。
【Abstract】 <Abstrcat> To investigate the effect of fresh hydrosalpinx fluid on the in vitro development of goat embryos, the fresh hydrosalpinx fluid was collected from estrus goat then added into media with different concentration for cultivating the eggs manipulated by microinjecting or nuclear transferring. The cleavage rate of zygotes incubate in medium containing 20% oviduct fluid (26.1%) was much higher than other groups containing 0 (7.6%), 50% (4.7%) and 100% (0) hydrosalpinx fluid in morula development. The zygotes injected with exogenous gene were cultured in the media containing 0, 20% and 50% hydrosalpinx fluid respectively. The proportion that developed into 16-cell stage was significantly higher (20.0%) at 20% hydrosalpinx fluid than that at other concentrations. There was no significant difference in morula development when cloned embryos cultured in 20% hydrosalpinx fluid media (7.3%) compared with those cultured in vivo (11.1%). In conclusion, the results indicate that the addition of 20% hydrosalpinx fluid to the in vitro culture media will enhance the 16-cell, morula development rate of zygotes, microinjection zygotes and cloned embryos.
