粒细胞集落刺激因子对大鼠颈动脉PCI后内膜修复和增生的影响

【摘要】 目的 探讨重组人粒细胞集落刺激因子(rhG CSF)对大鼠颈总动脉球囊损伤后再内皮化和内膜增生的影响。方法 5 6只Wistar大鼠随机分为2组。观察组2 8只:皮下注射rhG CSF30 μg·kg- 1 ·d- 1 。7d后行左颈总动脉球囊导管拉伤。术后1h、3、5、7、14d用扫描电镜、伊文氏蓝染色、HE染色、免疫组织化学方法,观察血管再内皮化、新生内膜增厚及细胞增殖情况。对照组2 8只:用生理盐水代替rhG CSF ,其余同观察组。结果 发现观察组的内膜再生面积(An)和An At(内膜损伤总面积)比值明显高于对照组(P <0 .0 1) ;内膜厚度(Ti)、Ti Tm(中膜厚度)比值及PCNA阳性细胞率均明显低于对照组(P<0 .0 1)。结论 干细胞动员剂rhG CSF显著促进球囊损伤后血管内皮化、抑制SMC和内皮细胞过度增生,从而抑制了球囊损伤后再狭窄的发生

【Abstract】 Objective To study the effects of mobilization agent of bone marrow stem cells, exogenous recombinant human granulocyte-colony stimulating factor (rhG-CSF), on the repair process of reendothelialization and neointima hyperplasy of rat carotid arteries injured with a balloon catheter. Methods A model of vascular restenosis was established on carotid arteries injured by a balloon catheter in 56 male Wistar rats randomized classified as rhG-CSF group and Contrast group.The animals were injected daily with 30 μg/kg rhG-CSF in rhG-CSF group or 0.9% NaCl in contrast group for 7 days. Respectively. On the eighth day after treatment, all animals underwent balloon angioplasty in common carotid artery. The animals were killed and the samples of carotid artery were harvested at 1 hour, 3rd,5th, 7th and 14th day respectively after balloon injury. The samples were studied with scanning electron microscopy (SEM),Evans blue staining. Morphometric analysis for endothelialization and. Immunohistochemistry for neointimal formation. The nuclear antigen(PCNA) were calculated for artery intima proliferation cell to evaluate the proliferation of vassel wall cells and the possible mechanism of the repair. of the injured carotid artery intima. Results SEM and Evan’s blue staining showed that th reendothelialization area (An) and the ratio of An/At (total area of the denuded vessels) in the rhG-CSF group were significantly more compared with that in control animals[(61.1±(4.52%) vs.39.2±4.36%,P<0.01).Neointima thickness was reduced by 38.8% in rhG-CSF-treated group compared with that in control group at 2rd weeks after injury. Immunohistochemical staining for PCNA positive cells were less in G-CSF-treated animals compared with that in control animals(42.6%vs.72.8%,P<0.01). Conclusions rhG-CSF has a beneficial effect on the reendothelialization and reduce neointima thickeness induced by the ballon injurey. Mobilization of EPCs by exogenous granulocyte colony stimulating factor may be a potential therapeutic strategy for prevention of restenosis after PCI.
