降钙素对体外培养的小鼠胚泡发育、分化的影响
Effect of calcitonin on development and differentiation of mouse blastocyst in vitro
【摘要】 目的:探讨降钙素(CT)与小鼠胚泡发育及植入的关系。方法:将孕D4小鼠胚泡随机分组并接种到添加不同成分的培养液内进行体外培养。于培养24、48h后在倒置显微镜下观察、记录胚泡的孵化、粘附及扩展生长情况。结果:培养24h后,添加10nmol/LCT组与联合培养组胚泡的粘附率分别为31.71%、32.35%,和对照组(16.30%)相比,均有明显增加。培养48h后,添加10nmol/LCT组与联合培养组胚泡的孵化率、粘附率、扩展率均比对照组有非常显著增高。结论:CT可明显地促进体外培养的小鼠胚泡孵化、粘附及扩展生长,与植入过程密切相关。
【Abstract】 Objective:To explore the effect of calcitonin (CT) on mouse embryo development and implantation. Methods: The mouse blastocysts were randomly divided into following groups: cultured with medium alone (served as control, groupⅠ), cultured with various calcitonin (groupⅡ, CT 5 nmol/L; groupⅢ, CT 10 nmol/L), and co-cultured with mouse endometrial cells (group Ⅳ). The rates of incubation, attachment and spreading of blastocysts in different groups were compared on 24 and 48 h after culturing.Results: After 24 h of culture, the attachment rates of blastocysts were 31.71% and 32.35% in group Ⅲ and group Ⅳ respectively; both were significantly higher than that in group Ⅰ (16.30%). After 48 h of culture, the percentages of incubation, attachment and spreading of blastocysts in groupⅢand groupⅣ were all significantly increased compared with the control. Conclusion:Calcitonin can improve the incubation, attachment and spreading of mouse blastocysts cultured in vitro.
