镀氮化钛钕铁硼对牙龈成纤维细胞表面形态及P70s6k表达的影响

【摘要】 目的观察牙龈成纤维细胞表面形态和细胞核糖体S6蛋白激酶(P70s6k)的表达,评价镀氮化钛钕铁硼磁体的生物相容性。方法将托槽、镀氮化钛及未镀膜钕铁硼磁体加入DMEM中制备成浸提液,以10%浓度加入培养的大鼠牙龈成纤维细胞,通过扫描电镜观察细胞形态的改变,免疫印记分析P70s6k的表达。结果加入托槽、镀氮化钛磁体浸出液培养的细胞表面形态良好、细胞膜完整,高倍扫描显示细胞膜表面及边缘有较多、较长的微绒毛,未镀膜磁体浸出液培养的细胞表面形态可见不光滑、不完整,细胞表面及边缘微绒毛短小。免疫印记结果显示,P70s6k表达的平均灰度值托槽组(76.0±1.50)与镀氮化钛磁体组(75.5±2.65)无显著差异(P>0.05)。未镀膜磁体组(40.4±1.75)与托槽组差异有显著性(P<0.01)。结论镀氮化钛钕铁硼磁体具有良好的生物相容性。

【Abstract】 Objective To observe the morphology of the gingival fibroblast and the expression of P70s6k to investigate the biocompatibility of magnet coated with titanium-nitride. Methods Brackets,magnet coated with titanium-nitride and bare magnets were incubated with DMEM to make solutions, 10% those solutions were added into cultured rat gingival fibroblasts, scan electron microscope was used to observe the morphology of cell membrane, western blotting was used to examine the expression of p70S6 kinase (P70s6k). Results The cells of bracket and titanium-nitride magnet groups were found with smooth surface, integrity membrane, more and longer microvillus on the surface and fringe of cells. The cells of bare magnet group were seen with coarse surface, incontinuity membrane,only a few short microvillus on the surface and fringe of cell. Western blotting showed that there was no significant difference for average gray values of P70s6k between bracket group (76.0±1.50) and titanium-nitride group(75.5±2.65,P>0.05),but there was significant difference between bared magnet group (40.4±1.75) and bracket group (P<0.01) . Conclusion Magnet coated titanium nitride has good biocompatibility.
