59Fe示踪谷氨酸包被三氧化二铁纳米颗粒的制备及其在小鼠体内的生物学分布
Determing the biodistribution of nano-Fe2O3-Glu in mice by 59Fe tracer and preparation
【摘要】 目的:应用59Fe对谷氨酸包被三氧化二铁纳米颗粒(nano Fe2O3Glu)进行示踪标记,研究其在小鼠体内的生物学分布。方法:采用59Fe标记谷氨酸修饰共沉淀法制备nano59Fe Fe2O3Glu,经肝间质或尾静脉注入小鼠体内,测定给药后不同时间nano59Fe Fe2O3Glu在血液、心、肝、脾、双肾、肺、脑、眼、性腺等主要组织器官中的放射性,观察其在动物体内的药代动力学特征,包括吸收、分布、消除的特点,组织蓄积及可能作用的靶器官。结果:nano59Fe Fe2O3Glu粒径15nm,59Fe标记的nano Fe2O3Glu经尾静脉注射,分别于1h和2天肝和脾达到高峰,峰值分别为48.9%ID/g和22.9%ID/g;肝间质注射后肝8天、脾24h达到高峰,在高、中、低三个剂量组肝脏分别为41.3%ID/g、20.8%ID/g和14.6%ID/g,脾分别为23.6%ID/g、21.2%ID/g和12.5%ID/g。脑、眼、性腺亦含有一定的放射性,表明纳米颗粒Fe2O3可通过血脑、血睾等屏障。经尾静脉和肝间质注射药物后,血液的放射性均在16天达高峰,然后逐渐下降,48天仍含有一定的放射性。结论:59Fe示踪nano Fe2O3Glu体内生物学分布灵敏、准确,其在体内主要的靶器官是肝和脾,无血液毒性与细胞毒性。
【Abstract】 Objective:To observe the distribution of nano-particles iron oxide in mice ,which is coated with Glu labeled by 59Fe (nano- 59Fe-Fe 2O 3-Glu). Methods:By coprecipitation to synthesis nano- 59Fe- Fe 2O 3-Glu,the distribution in blood, heart, liver, spleen, kidneys ,lungs, brain, eyes and gonad etc are measured, and the character of its pharmacokinetics in vivo is observed,such as the behavior of the absorption, distribution and clearance, accumulation in tissues and the probable target-organ. Results:Nano- 59Fe-Fe 2O 3-Glu,the diameter of which is 15 nm, the maximum in the liver is 48.9%ID/g and the maximum in the spleen 22.9%ID/g at 1 h and 2d respectively after injection of vein of nano- 59Fe-Fe 2O 3-Glu , while in the group of injection of inter-liver the maximum in the liver is 41.3%ID/g、20.8%ID/g、14.6%ID/g at 8 d and the maxium in the spleen is 23.6%ID/g、21.2%ID/g、12.5%ID/g at 24 h in high dose, mid-dose and low dose respectively.There is some radiation in brain ,eyes and gonad ,which indicates nano-Fe 2O 3 may pass through blood brain barrier ,blood testis barrier.The radiation in the blood increases and attains the maximum at 16 d both in the group of injection of vein and in the group of injection of inter-liver, and then it decreased ,but it can also be observed at 48 d. Conclusion:The biological distribution of nano-Fe 2O 3-Glu,which is mainly detained in liver and spleen in vivo, is traced by 59Fe sensitively and accurately, without hemotoxicity and cytotoxicity.
