大肠杆菌流行株耐药特性对其免疫效果的影响
Effect of Resistance on Immunity of Epidemic Escherichia coli
【摘要】 经人工诱导不同耐药水平的菌株,改变其耐药特性,制成灭活制剂免疫小鼠,2周后分别用致死剂量的原菌株和24株同期分离菌株感染免疫小鼠,考察小鼠存活率。结果表明:“O”抗原相同的菌株免疫小鼠,耐药种类少且耐药水平较低的1号菌株对24株同期分离的致病性鸡大肠杆菌的保护率仅为31%,免疫后接受体外敏感药物环丙沙星(CIP)治疗保护率为34%。经体外诱导耐药后保护率无明显变化,但再给药治疗保护率明显提高,达到54%。而135号菌株对受试的11种抗生素耐受9种,且耐药水平较高,其培养物免疫小鼠后对致病大肠杆菌感染的保护率显著提高,达到67%,含有抗生素培养基培养的菌液免疫小鼠,保护率略高于普通培养时的保护率,达到72%。当使用体外不敏感药物CIP对免疫小鼠治疗时,其免疫保护率达到最高77%,但使用抑制剂抑制其耐药性后,保护率随之大幅下降,为37%。7号菌耐受6种抗生素耐药水平中等,无药培养后灭活物的保护率为54%,含药培养后灭活物的保护率为61%,体外不敏感药物在体内可以加强免疫保护作用,保护率达到79%。大肠杆菌流行菌株的耐药特性可能与其免疫学效果有关,多重耐药株对当前流行菌株具有更好的免疫保护作用,而且已耐受抗生素可加强免疫保护作用。
【Abstract】 To understand the immunologic effect of Escherichia coli with different resistance,some strains were induced with drugs or treated with inhibitor in order to change their resistance.Two weeks after the selected strains were vaccinated with inactivated cultures(at 37℃ for 18 h),KM mice were injected intraperitoneally with wild pathogenic strains.Then survival rates of the mice were detected within a week.The survival rate of mice immunized with strain 1 which had lower resistance level and narrower pattern to drugs was only 31% when injected with 24 strains of wild pathogenic E.coli.When the mice were treated with CIP at the same time,the rate was 34%.When strain 1 was induced to resist more drugs,the rate went up to 54% remarkably.But the survival rate of mice immunized with inactivated cultures in Luria-Bertani(LB) broth of strain 135 which was resistant to 9 out of 11 kinds of drugs used in the test and had higher resistance level was 67% after the mice were injected with 24 strains wild pathogenic E.coli.When CIP(1/2MIC) was added to the medium during strain growth,the rate was 72%.When the mice were treated with CIP at the same time the rate went up to 77%.Furthermore,the rate went down to 37% after some resistance of strain 135 was inhibited partly.Strain 7 was also a multi-resistant one.When it was cultured in LB,or in LB with CIP,or treated with CIP,the rates were 54%,61% and 79% respectively.Immunologic effect of epidemic E.coli may be associated with drug resistance.Multi-resistant strains were more effective on immunologic protection,which was enhanced by CIP(resistant,in vitro) in vivo.
