联合STZ和ALX制备大鼠糖尿病模型的心肌病理学观察
Pathological changes on myocardium of STZ and ALX-induced diabetic model in Wistar rats
【摘要】 目的 联合STZ和ALX制备大鼠糖尿病模型 ,观察其心肌组织病理学改变。方法 选取Wistar大鼠 30只 ,随机分两组 ,应用STZ(30mg/kg)和ALX(5 0mg/kg)对模型大鼠分别一次性腹腔注射 ,常规法测定血糖血脂 ,光镜及电镜观察其心肌病理学改变。结果 给药 3天后模型组血糖值即≥ 16 .5mmol/L的标准值为 (2 2 .6 5± 3.0 2 )mmol/L ,对照组 (4 .96± 0 .39)mmol/L ,P <0 .0 5 ;模型组TG(1.6 1± 0 .13)mmol/L ,对照组 (1.2 1± 0 .2 4 )mmol/L ,VLDL(32± 0 .0 3)mmol/L和 (0 .2 4± 0 .0 3)mmol/L ,P <0 .0 5 ;心脏重量 /体重模型组 (4 .10± 0 .2 8) ,对照组 (2 .4 0± 0 .17) ,P <0 .0 5。药物模型组光镜下 (HE染色 )心肌排列紊乱 ,细胞变性、坏死 ;电镜下可见心肌内细胞器病变。模型鼠糖尿病症状明显。结论 此方法制备的糖尿病大鼠模型经济实用 ,简便易行 ,可用于糖尿病心肌病的发病机制及药物干预方面的研究
【Abstract】 Objective To observe the pathological changes on myocardium by establishing a STZ and ALX-induced diabetic model in Wistar rats.Methods Thirty Wistar rats were divided into 2 groups randomly.The model rats were injected with STZ(30mg/kg) and ALX(50 mg/kg) in left abdominal cavities .Blood glycose,blood lipid were cheked before optical and electron microscope observation on pathological changes of myocardium.Results In model rats,blood glycose rose up 3 days after injection and remained over 16.5mmol/L in 12th weeks during the whole experimental periods,significantly higher compared with the control group〔(22.65±3.02)mmol/L vs (4.96±0.39)mmol/L,P<0.05〕.There were also distinct elevation in blood lipids 〔TG(1.61±0.13)mmol/L vs (1.21±0.24)mmol/L;VLDL(32±0.03)mmol/L vs (0.24±0.03)mmol/L,P<0.05〕 and HW/BW(4.10±0.28 vs 2.40±0.17,P<0.05).The evidences on optical microscope and electron microscope confirmed pathological changes on myocardium of diabetic models along with the diabetic symptoms (polydipsia,polyphagia,diuresis).Conclusion We believe that little dosage STZ and ALX-induced diabetic model in Wistar rats are cost-efficient and easy to prepare.The model could be used to study on pathogenesis and medicinal interference on diabetic cardiomyopathy.
