【摘要】 目的建立测定血浆中黄芩苷的高效液相色谱法,研究黄芩苷在大鼠体内的药物动力学行为。方法大鼠经灌服清热合剂后,血浆样品经甲醇沉淀蛋白后,进行色谱分离,测定其血浆中黄芩苷的含量。结果黄芩苷最低定量浓度为0.025μg·ml-1,线性范围为0.025～2.5μg·ml-1,精密度与准确度符合生物样品分析要求。动物实验结果显示,黄芩苷在大鼠体内出现多峰现象。结论所建立的方法简便快速,适用于黄芩苷的血药浓度测定及药物动力学研究。更多还原

【Abstract】 OBJECTIVE To establish an RP-HPLC method for determination of baicalin in plasma and study pharmacokinetics of baicalin in rats in vivo. METHODS Plasma pretreated with methanol precipitation was performed chromatographic separation and baicalin concentration in rat plasma samples after intragastric administration of qing re mixture was detected. RESULTS The linear range of baicalin was 0.025-2.5 μg·ml~-1 and the lower limit of quantitation(LLOQ) was 0.025 μg·ml~-1 ,with accuracy and precision conformed with the requirement of biological specimen analysis. The results of animal experiment showed that the baicalin in QRHJ presented multiple-peak phenomenon in the body of Wistar rats. CONCLUSION The established method is simple, rapid and suitable for the determination of plasma concentration of baicalin and the study of pharmacokinetics.更多还原
