发酵培养中葡萄糖对超螺旋质粒DNA产量的影响

【摘要】 本文首先研究了含有不同质量浓度葡萄糖的发酵培养基对超螺旋质粒DNA产量的影响。结果表明,发酵培养基中最佳葡萄糖的质量浓度为2 00g L。此培养基与未添加葡萄糖的培养基相比,超螺旋质粒DNA产量提高2 56倍;菌体密度提高1 26倍。其次研究了优化培养基下影响超螺旋质粒DNA产量的各参数变化规律。

【Abstract】 In this paper the influence of glucose on the production of supercoiled plasmid DNA in the fermentation process is presented.Glucose of different concentration (0 g/L,1.00 g/L,2.00 g/L,4.00 g/L) was used as carbon source.The optimal concentration of glucose in culture medium was 2.00 g/L. Supercoiled plasmid DNA concentration was enhanced by 2.56 and biomass concentration was enhanced by 1.26 in comparison with those without glucose supplement in the optimized culture medium.Factors influencing the production of supercoiled plasmid DNA were studied.
