AccQ·Tag法测定强化食品中的牛磺酸
DETERMINATION OF TAURINE IN ADDED FOODS BY AccQ·Tag METHOD
【摘要】 食品中强化的牛磺酸经沉淀蛋白质或直接水溶提取后 ,采用Waters公司提供的AccQ·Fluor试剂柱前衍生后 ,利用反相高效液相色谱法测定。使用AccQ·TagC18柱 ,以pH =4 .9的 0 .0 2moL/L磷酸二氢钾 -乙腈 -水 (体积比为 85∶9∶6 )为流动相 ,流速为 1.0mL/min ,在 2 4 8nm紫外检测。牛磺酸的线性范围为 5~ 2 0 0 μg/mL ,相关系数r =0 .9991,RSD为 0 .5 9%~ 1.9% ,回收率为 97.2 %~ 10 1.9%。
【Abstract】 Taurine in added foods was extracted by depositing protein or being dissolved in water, and then it was determined on AccQ·Tag C 18 column by pre column derivatization RP-HPLC. AccQ·Fluor reagent manufactured by Waters corporation was used as the derivative reagent. The mobile phase is 0.02 moL/L KH 2 PO 4 solution (pH=4.9)-acetonitrile-water (85∶9∶6), the flow rate is 1.0 mL/min and the detection wavelength is 248 nm. The linear range is 5~200 μg/mL with the correlative coefficient of 0.999 1, RSD is 0.59%~1.9% and the recovery is 97.2%~101.9%.
