电位型无标记IgG免疫探针测人IgG
Determination of IgG in human serum by the potentimetric no sign IgG immunoprobe
【摘要】 以 Pt 为基体电极,用戊二醛交联法在 Pt 电极上固定兔抗人 IgG 抗体,制成 IgG 免疫探针。根据人免疫球蛋白(IgG)能与兔抗人 IgG 抗体发生定量的特异性免疫反应,利用电化学方法,找到了测定人血清中 IgG的简单方法。人 IgG 的含量在0.2~1.2μg/L 之间,与兔抗人 IgG 免疫探针的电位变化值(△E)呈良好的线性关系。工作曲线的回归方程△E=23.0 c+9.733,相关系数 r=0.9986,检测下限小于0.2μg/L。用于正常人血清中 IgG 含量的测定,加标回收率为95%~103%,结果满意。
【Abstract】 On Pt electrode,the specific immunological reaction between the human immunoglobulin G (IgG) and the rabbit and human antibody can be carrid into practice quantitatively.Using electchemisty theory,a covenient method for determination of the IgG has been found.The calibration curve for hIgG was obtained.In the concentration range of 0.20 to 1.2 μg/L with the correlation coefficient of 0.998 6.The method was used for the determination of IgG in normal human serum with good satisfactory results.The analytical recoveries are 95%、96%、103% for three replicates.
