氧化低密度脂蛋白对血管平滑肌细胞凋亡的影响
Study on the effects of ox-LDL on the cultured human SMC
【摘要】 目的:探讨氧化型低密度脂蛋白(Oxidizedlowdensitylipoprotein,oxLDL)对人血管平滑肌细胞(Smoothmusclecell,SMC)凋亡的影响。方法:利用密度梯度超速离心分离纯化人血浆LDL,体外和CuS04共孵育氧化生成oxLDL。不同浓度的oxLDL对SMC作用不同的时间,观察其对SMC凋亡状况的影响。结果:oxLDL对SMC的作用具有时间浓度依赖性,较低浓度的oxLDL促SMC增生和泡沫细胞化,高浓度时则诱导SMC凋亡发生。结论:适当浓度的oxLDL促SMC增生及表型转换而高浓度时则对SMC促凋亡作用。
【Abstract】 Objective: Study the effects of ox-LDL (oxidized low density Lipoprotein) on the apoptosis of cultured human SMC(smooth Muscle cell).Methods: Through NaBr density gradient centrifugation, LDL was isolated and purified from human plasma, ox-LDL was produced from LDL by being incubated with cuso4, ox-LDL was than added to the medium at different concentration for different time. The effects of ox-LDL on SMC apoptosis was investigated from different aspects.Results: ox-LDL can promote proliferation and foam-cell producing of SMC at lower concentration and induce SMC apoptosis at higher concentration. Conclusion: ox-LDL played an important role in SMC apoptosis.
