杜氏盐藻硝酸盐还原酶缺陷型突变藻株的分离和初步鉴定

【摘要】 目的:分离制备杜氏盐藻硝酸盐还原酶(NR)缺陷型突变藻株并进行初步鉴定。方法:①杜氏盐藻经乙 基亚硝基脲(ENU)诱变后,利用氯酸盐抗性筛选方法获得氯酸盐抗性藻株,经复筛和不同氮源生长实验,获得NR 缺陷型突变藻株。②用磺胺比色法测定酶反应生成的亚硝酸盐(NO-2)的浓度,从而计算出盐藻NR活性。比较突 变藻株与野生型杜氏盐藻NR活性,作出初步鉴定。结果与结论:共获得30株NR缺陷型突变藻株,共有6株NR 活性显著降低。杜氏盐藻NR缺陷型突变藻株成功建立。

【Abstract】 Aim:To isolate and identify nitrate reductase(NR)-deficient mutants of Dunaliella salina (D.salina).Methods: After ethynitrosourea mutagenization, chlorate-resistant mutants were acquired by chlorate-resistant selection. By reselection and growing experiments with different nitrogen cultures, NR-deficient mutants were got. The concentration of nitrite produced in enzyme reaction was determined by sulfanilamide colormetry and then NR activity was calculated. Comparing NR activity of NR-deficient mutants with wild type D. salina, the mutants were identified. Results and Conclusion:Thirty NR-deficient mutants were obtained,and NR activity of 6 mutants decreased significantly. NR-deficient mutants of D.salina were established successfully.
