犬肺气肿模型的制作
Establishment of pulmonary emphysema models with dogs
【摘要】 目的:探讨大动物肺气肿模型制作方法。方法:选 16只健康杂种犬,随机均分为 2组。应用木瓜蛋白酶行气管内注入及雾化吸入 2种方法,呼吸机过度通气诱导,每种方法分单、双侧肺 2种情况 (各 4只犬 )。诱导后1~3周行大体标本观察,测量肺体积,病理学检查肺组织结构。结果: 2种方法所制的肺气肿模型肺组织表面均可见肺大泡形成,病理学检查:肺组织结构破坏,肺泡融合成肺大泡。模型肺体积与正常肺体积比较差异有统计学意义(P<0. 001)。雾化吸入法各叶肺组织呈均质型肺气肿改变,注射法各叶肺组织呈非均质型肺气肿改变。双腔气管插管单侧用药,用药侧肺组织呈均质型及非均质型肺气肿改变,对侧肺组织结构正常。结论:木瓜蛋白酶加呼吸机过度通气,能快速诱导大动物肺气肿模型。不同方法能够诱导出不同类型肺气肿模型。
【Abstract】 Aim: To probe into the methods of establishing pulmonary emphysema models with big animals. Methods: Sixteen healthy mongrel dogs were chosen. Chinese flowering quince proteinase was given by intratracheal injection or atomization absorbing.Using respirator to induce excessive ventilation. Experiments were performed on single lung and both lungs for each method.After 1~3 weeks the whole specimens were observed,the pulmonary volume was measured, and the pulmonary tissue underwent pathological examination. Results: Pulmonary alveolus could be seen on the surface of pulmonary tissue of the models made by the two methods. Pathological examination showed damages of pulmonary tissue. Compared with normal pulmonary volume, model pulmonary volume was obviously bigger (P<0.001). For the models made by atomization absorbing method, the tissues of each lung appeared evenly-spreaded, and for the models made by injection method, the tissue appeared non-evenly-spreaded. For the models intubated the double cavities and applied drugs in one side, the lung tissue in which side drugs were applied appeared evenly-spreaded and non-evenly-spreaded changes. The structure of the other side was normal. Conclusion: Chinese flowering quince proteinase plus excessive ventilation of the respirator can rapidly induce pulmonary emphysema models with dogs. Different methods can induce different models for related research.
