小鼠生长过程中肝AKP的活性变化及细胞定位
Activity Changes of Alkaline Phosphatases and Its Histo-Cellular Locatin in Mouse Liver during Growth
【摘要】 用比色法、酶的原位复性电泳、组织化学和细胞化学方法等分析观察了小鼠生长过程中肝碱性磷酸酶(Alkaline phosphatase,AKP)同工酶变化和细胞定位.比色法和细胞化学法研究表明,在小鼠生长过程中,出生第 7天小鼠(D7)肝的AKP活性最强,成体肝(Da)活性最弱; AKP活性在肝小叶内带、中带、外带中的活性呈依次减弱的趋势;细胞化学显示,AKP活性主要出现在肝实质细胞的细胞膜上,尤以胆小管周围的细胞膜上最多.用酶的原位复性电泳测得小鼠生长过程中共出现四条AKP同工酶带,各同工酶活性随着小鼠的生长发育时间不同而发生变化.
【Abstract】 In this study we observed the activity changes of alkaline phosphatase(AKP) and its histo-cellular location in mouse liver during growth. By colorimetry and histo-chemical method we found the activity of AKP ,which was highest in D7 and lowest in Da,ascended first followed by decline. Four isoenzyme of AKP was determined by renaturation electrophoresis. The activity of each isoenzyme changed differently from other isoenzyme during the mouse’s growth. The results of cellular-chemistry showed that the activity of AKP is mainly located in the membrane of liver cell especially more in the zone of canaliculi.
