瓜叶菊花梗和花托的愈伤组织诱导与植株再生研究
In Vitro Culture and Plant Regeneration of Pedicel and Receptacle of Senecio×hybridus
【摘要】 为建立瓜叶菊愈伤组织再生体系,以瓜叶菊花梗、花托为外植体进行了愈伤组织诱导和分化研究.结果表明:1)在MS+2,4-D2mg/L+NAA0.1mg/L,3/4MS+2,4-D2mg/L+6-BA1mg/L+NAA0.1mg/L,3/4MS+6-BA2mg/L+NAA0.1mg/L培养基上花梗、花托的出愈率可达100%,但在3/4MS+6-BA1mg/L+NAA0.1mg/L,3/4MS+6-BA1mg/L培养基上花梗的出愈率不及花托;2)3/4MS无激素培养基对花托愈伤组织芽的分化较好,诱导率可达95%,3/4MS+6-BA1mg/L培养基对花梗愈伤组织芽的分化较好,诱导率为90%;3)再生苗在1/2MS+IBA0.2mg/L培养基中生根完成植株再生.
【Abstract】 In order to establish the plant regeneration system of callus of Senecio×hybridus,using pedicel and receptacle as explants and taking MS as basic media,callus induction and differentiation of Senecio×hybridus were studied.The result show that:1) The media of MS + 2,4-D 2 mg/L + NAA 0.1 mg/L,3/4 MS +2,4-D 2 mg/L +6-BA 1 mg/L +0.1 mg/L NAA,3/4 MS +6-BA 2 mg/L + NAA 0.1 mg/L were optimum to induce callus from pedicel and receptacle with a rate of 100% frequency;while,on the media of 3/4 MS + 6-BA 1 mg/L + NAA 0.1 mg/L,3/4 MS + 6-BA 1 mg/L,the rate of callus inducement from pedicel was lower than that from receptacle;2) The differentiation media of 3/4MS without plant growth regulator was optimal to induce buds from callus of receptacle and had a 95% plant regeneration,the differentiation media of 3/4MS + 6-BA 1 mg/L was optimal to induce buds from callus of pedicel and had a 90% plant regeneration;3) The seeding on the rooting media of 1/2MS + IBA 0.2 mg/L produced roots and turned out a whole plant on the 10th day.
